| 产品参数 | 概述托珠单抗是日本中外制药公司研发的并于2009年在欧盟上市的重组人IL-6单克隆抗体，可与可溶解型和膜结合型IL-6受体(sIL-6R和mIL-6R)特异性结合，抑制IL-6介导的信号转导。托珠单抗于2008年4月、2009年1月和2010年1月先后在日本、欧洲和美国获准用于治疗RA。目前，托珠单抗已进入中国市场。2013年7月，我国专家提出了《托珠单抗治疗类风湿关节炎的专家建议》。近年来，托珠单抗在AOSD治疗中逐渐显示出良好的有效性和安全性。
托珠单抗与DMARD联用在1220例活动期RA患者参与的多中心随机双盲安慰剂对照TOWARD研究中，患者在服用稳定剂量DMARD的基础上分别静脉注射8mg/kg托珠单抗或安慰剂。经24周治疗后发现，托珠单抗＋DMARD组达到ACR20、ACR50和ACR70的比率为60.8%、37.6%和20.5%，DAS＜2.6的比率为30%，安慰剂＋DMARD组达到ACR20、ACR50和ACR70的比率为24.5%、9.0%和2.9%，DAS＜2.6的比率为3%。与单用DMARD的患者相比，接受联合治疗的患者在达到ACR20、ACR50的比例和8处关节疾病活动度评分(DAS8)方面均有明显优势。认为托珠单抗联合任一种DMARD均可使RA患者在无明显不良反应情况下获得安全有效的全身系统和关节局部的症状缓解。 |
